CDH13geneencodesamemberofthecadherinsuperfamily.TheencodedproteinislocalizedtothesurfaceofthecellmembraneandisanchoredbyaGPImoiety,ratherthanbyatransmembranedomain.Theproteinlacksthecytoplasmicdomaincharacteristicofothercadherins,andsoisnotthoughttobeacell-celladhesionglycoprotein.Thisproteinactsasanegativeregulatorofaxongrowthduringneuraldifferentiation.Italsoprotectsvascularendothelialcellsfromapoptosisduetooxidativestress,andisassociatedwithresistancetoatherosclerosis.Thegeneishypermethylatedinmanytypesofcancer.
Full-lengthextracellulardomainofhumanCDH13gene(139-693aa)wasconstructedwith31N-terminalT7/HistagandexpressedinE.coliasinclusionbodies.Thefinalproductwasrefoldedusingourunique“temperatureshiftinclusionbodyrefolding”technologyandchromatographicallypurified.
