CytoForm®-200consistsofa4.5x4.5x0.15cm3DprintedscaffoldmadeoutofHyperelasticBone™(trademarkpropertyofDimensionInx®).ItcomesasalargesheetthatcanbereADIlycutorpunchedouttomeetexperimentalneeds.
TheHyperelasticBone™scaffoldsconsistof~90%Hydroxyapatiteintheformof1-30µmsphericalparticles,and~10%polylactide-co-glycolide(PLG).
Thesescaffoldsprovideinterconnectedporosity(forvascularization),highbioactivity,easyhandling,workABIlityandhighmechanicalstability.Thesefeaturesmakethesubstratesidealforcellcultureinboneandhardtissueregeneration.
TheCytoForm®-200scaffoldsareethanolwashedpriortoshipment,butarenotconsideredsterile.
EveryotherlayerisoffsetintheXandYby0.5mmtoprovideoptimalcell-seedingsurfacearea.
CytoForm®-200ischemicallystableinneutralpH,isflexIBLe,butalsohascompressive/mechanicalstrength.
ScaffoldscanbeseededwithcellsorcellscanalsobesUSPendedinapre-hydrogelmaterial(suchasTypeIcollagen).

Sterilization
Itisrecommendedtocutorpunchsamplesintendedfortheexperimentpriortosterilization(ie.4mmbiopsypunch).
- Washsamplesin70%ethanolfor30-60minutes.
- Rinsesamples3-5timesinsterilewaterorPBS.
- Removeexcessliquidfromscaffoldsviadabbingonfilterpaperorsterilizecloth,oraspiratingoutexcessliquid.Donotallowscaffoldstobecomecompletelydrypriortocellseeding.
CellSeedingInstructions
- Placewettedscaffoldsintodesiredtissuecultureplate.
- Dispense10uLofcellsuspensionontothescaffoldfora4-5mmdiameterscaffold(~50,000cells).
- Allowcellstoattachfor30-60minutespriortoaddingadditionalmedia.
- Slowlyfillthecellculturewellwithmediaandincubateat37°C
CellSeedingInstructionsUsingCollagenHydrogel
- Placewettedscaffoldsintodesiredtissuecultureplate.
- Addcellstoaneutralizedcollagensolution(suchasPureCol®EZGel).
- Slowlydispensethecollagen/cellsolutionoverthescaffoldandallowthesolutiontofullypermeatethroughoutthescaffold.
- Allowcollagentopolymerizewithinthescaffoldfor60-90minutesat37°C.
- Fillthewellwithcellculturemediumuntilthescaffoldiscompletelycoveredandcontinueincubatingat37°C.]
