IRGACURE2959isahighlyefficientnon-yellowingrADIcalphotoinitiatorfortheUVcuringofsystems,suchashydrogelsandbioinks.Irgacureismostefficientlyactivatedusing365nmwavelengthUVlight.Irgacuremustbesolubilizedinmethanol,asitisnotwatersoluble(suchasLAPandRuthenium).
Thisirgacureproductisconsiderednon-sterile.Addingantibioticstoyourcellculturesystem,orsterilefilteringisrecommended.Tosterilefilter,resUSPendthedesiredquantityofirgacureandfilterthroughsmall0.2micronbuttonfilters.Usethesterilephotoinitiatorwithin2weeks.Thisproductcomeswith5gramsofdrypowder.

Theproductshipsongelpacks.Storetheproductat2-8°C.Weighouttherequiredamountofpowdertosolubilize.Oncesolubilized,usewithin2weeks.

DirectionsforUse
- Tosolubilizethephotoinitiator,solubilizeirgacureinneatmethanolina10%solution(100mg/ml).Note:Thephotoinitiatorinsolutionhasashelflifeof2weeks.Onlydissolverequiredamountofphotoinitiatorrequired.Storeremainingphotoinitiator(powderorsolution)at2to10°C.
- Calculatethevolumeofthephotoinitiatorrequiredbymultiplyingthetotalvolumeofsolution(ie.methacrylatedcollagen)requiredby0.01.Forexample,ifyouaremaking10mlofcollagenmethacrylate,thecalculatedvolumeofthephotoinitiatortoaddis100µl.
- Addthecalculatedvolumeofphotoinitiatortotherequiredvolumeofcrosslinkablesolutionandmixthoroughly.
- Addyourcellstothepre-hydrogel/photoinitiatorsolution.
- Dispenseorbioprintyourhydrogel/photoinitiator/cellsolutionintothedesiredcultureware(i.e.6-wellplate,48-wellplate).
- ForUV-crosslinking,placethepre-hydrogel/printedstructuredirectlyundera365nmUVlightcrosslinkingsource.Note:Longerexposureallowsmorecrosslinking,thougheachcelltypewithstandsdifferentdegreesofUVlightandfreeradicalexposure(generatedbythephotoinitiator)thatmediatescrosslinking.
Youmaytunephotoinitiatorconcentration,lightintensityandPhotocrosslinkingtimetocustomizefinalhydrogelstiffness
